Leveraging synthetic biology to create high-value compounds inaccessible by conventional chemistry
Throughout the 20th century, chemical synthesis (chemistry) laid the foundation for human prosperity, delivering transformative innovations such as fertilizers, plastics, and life-saving pharmaceuticals. However, modern society now faces an unprecedented era of challenges—from climate change and emerging infectious diseases to geopolitical risks that threaten supply chain stability and economic security. Our current manufacturing models, heavily reliant on fossil resources and centralized global supply chains, are reaching their structural limits.

The Yuzawa Lab confronts these critical issues through a synthetic biology approach that transcends the constraints of traditional chemistry. Leveraging advanced long-chain DNA synthesis and editing technologies, we precisely engineer megasynthases and microbes to establish production systems for a diverse array of high-value molecules (MW 100–1000). These compounds, which are otherwise inaccessible or prohibitively difficult to synthesize chemically, can now be produced through sustainable and self-reliant processes.

By establishing advanced molecular manufacturing as a core strength, we aim to provide a versatile toolkit for the autonomous creation of complex functional molecules. Through these efforts, the Yuzawa Lab contributes to the realization of a resilient and prosperous society that remains steadfast even in an uncertain world.

RESEARCH
The mission of the Yuzawa Laboratory is to create high-value compounds that cannot be achieved through conventional chemistry by leveraging synthetic biology. Traditional organic synthesis often faces significant challenges in efficiently producing large, structurally complex molecules containing multiple stereocenters. In contrast, synthetic biology harnesses the remarkable catalytic precision of biological systems, enabling the production of structurally diverse and highly stereochemically defined molecules with molecular weights ranging from approximately 100 to 1,000.

Our laboratory engineers and redesigns megasynthases, including polyketide synthases (PKSs) and nonribosomal peptide synthetases (NRPSs), and precisely integrates their corresponding biosynthetic gene clusters into microbial genomes to establish innovative biosynthetic pathways. This engineered megasynthase platform not only enables the production of novel high-value compounds but also provides a sustainable approach for manufacturing commodity chemicals from renewable feedstocks.

By integrating our proprietary long DNA synthesis and editing technologies, for which patent applications have been filed, with advanced megasynthase engineering, together with the biofoundry expertise and experience cultivated in the United States, the Yuzawa Laboratory aims to establish next-generation microbial production platforms. Through these efforts, we seek to create molecular architectures that are inaccessible through conventional approaches while advancing sustainable biomanufacturing for a future bio-based society.

The Institute for Advanced Biosciences at Keio University is known for one of the world's leading research facilities for metabolome analysis and provides a wide variety of research equipment that enables advanced R&D and technology demonstration.

In synthetic biology research, we design and build genetically engineered microbes and then test them by measuring their metabolomes. This Design-Build-Test cycle gives us big data to deeply interpret our enzyme/microbe designs (= Learn) by machine-learning etc.
